Cell Counting Kit-8 (CCK-8) is a highly sensitive, non-radioactive reagent designed for rapid and accurate quantification of cell viability, cell proliferation, and cytotoxicity. Using a water-soluble tetrazolium salt (WST-8), CCK-8 provides a simple colorimetric assay based on dehydrogenase activity in live cells. The resulting formazan dye correlates directly with the number of metabolically active cells and can be measured via absorbance at 450 nm.

This assay eliminates the need for radioisotopes or solubilization steps and is compatible with high-throughput formats. CCK-8 offers exceptional sensitivity, minimal cytotoxicity, and reliable results across a broad range of cell types.

Technical Notes
CCK-8 detects dehydrogenase activity in live cells. Chemicals that alter mitochondrial function may affect the assay.

Avoid introducing bubbles into the wells, as this may interfere with optical density readings.

If reducing agents are present, monitor and subtract background absorbance, as WST-8 may be reduced non-enzymatically.

Incubation times may vary by cell type. Leukocytes or low-density cultures may require extended incubation.

For turbid samples, subtract the background O.D. at 600 nm or higher to correct the signal.

If sterilization is needed, filter the reagent using a 0.2 µm membrane.
